Almost every polite refusal at work follows one simple pattern. Use this template every time:
What each part does · 各パーツの役割
- Soft opener — "I'm sorry, but…", "Unfortunately…"
これから難しいことを伝える、と相手に知らせます。「No」が唐突に感じられなくなります。 - Reason — a short sentence explaining why.
断りを理解してもらうのに十分な文脈を与えます。理由は一文で十分です。 - Alternative (optional) — "Maybe I can… instead."

Linguistic Nuance · 細かなニュアンスの解説
"No, I can't." with nothing around it sounds cold, even if that's not your intention (前後にクッションとなる言葉がないと冷たく聞こえます)。
"I'm sorry, but I can't take this on right now." tells the listener something difficult is coming, so the refusal feels considered rather than sudden (難しい話が来ることを先に伝えるだけで、印象が大きく変わります)。
